PaceSetter Application Notes With the recent FDA allowance of whole slide imaging for the review and interpretation of digital surgical pathology slides, as well as growing quantitative biomarker analysis of tissue sections, histology labs are increasingly standardizing the process of preparing slides. Standardization of all steps of the histology workflow, including embedding, processing, microtomy, and staining, can impact slide quality and consistency both within and across labs.

PaceSetter Section Mounting System with Microm Microtome

PaceSetter Section Mounting System paired with a Thermo Scientific™ (Microm) HM 355S microtome. The PaceSetter can also be paired with Leica motorized microtomes.

The PaceSetter section mounting system is the first benchtop system to provide standardization and improved section quality across all stages of microtomy – cutting, floating, and mounting tissues. The PaceSetter interfaces with motorized microtomes and utilizes a gentle water stream to transfer paraffin-embedded sections from the microtome blade to slides. Sections are exposed to warm water for relaxing at a temperature and for an amount of time defined by the user. Users can also program the number of sections per slide, the location of section placement on the slide, and whether sections are mounted serially or with different levels on the same slide.

Improved Slide Quality
The PaceSetter provides provides standardized and consistent slide quality in the following ways:

  • Consistent section thickness: By interfacing with motorized microtomes and making sectioning hands-free, the PaceSetter encourages the using motorized microtomy. Utilizing motorized microtomy, versus manually turning the advance wheel, is known to offer more consistent section thickness than manual use of the same microtome. Section thickness impacts stain quality (Chlipala et al) and can affect diagnosis.

  • More useable sections: Gentle and hands-free section transfer reduces the damage of sections by manual transfer to the water bath, yielding more useable sections.
  • Programmable and reproducible section floating: The PaceSetter allows the user to set the temperature and time for section floating. These conditions can be standardized within and across laboratories, and provides better control over mounted section thickness, width, and length over manual floating and mounting methods.
  • Precise section placement: The PaceSetter provides consistent section placement across all slides, at the top, middle, or bottom of the slide, whether one or two sections are placed on each slide. As well, the robotic and hands-free handling of slides removes any inconsistencies in section mounting (e.g., stretch from slower removal from the water bath) that may occur during manual section mounting.
